Molarity of Normal Saline
Normal saline is 0.9% w/v sodium chloride — that works out to 0.154 mol/L.
At a glance
The Short Answer
Normal saline is a 0.9% weight-per-volume NaCl solution, meaning 9 g of sodium chloride per liter. Dividing by the molecular weight of NaCl (58.44 g/mol) gives 9 ÷ 58.44 = 0.154 M, or 154 millimolar (mM). That is the molarity used in IV fluids and lab buffers everywhere.

The Math, Step by Step
- Convert the percentage: 0.9% w/v = 0.9 g per 100 mL = 9 g per 1,000 mL.
- Convert grams to moles: 9 g ÷ 58.44 g/mol = 0.1540 mol.
- Divide by liters: 0.1540 mol ÷ 1 L = 0.154 M.
NaCl Content by Bag Size
| Bag Volume | NaCl Mass | Moles of NaCl | Molarity |
|---|---|---|---|
| 250 mL | 2.25 g | 0.0385 mol | 0.154 M |
| 500 mL | 4.50 g | 0.0770 mol | 0.154 M |
| 1,000 mL | 9.00 g | 0.1540 mol | 0.154 M |
Why 0.154 M Is "Normal"
NaCl dissociates into Na+ and Cl− ions, so a 0.154 M solution delivers roughly 2 × 154 = 308 mOsm/L of dissolved particles. That closely matches the osmolarity of blood plasma (about 275–295 mOsm/L), which is why normal saline is considered isotonic and safe for intravenous use.
Making Your Own
To prepare 1 L of 0.154 M NaCl in the lab, weigh 9.0 g of sodium chloride, dissolve it in about 800 mL of distilled water, and dilute to a final volume of exactly 1 L. For sterile medical use, only commercially prepared saline should be used.
Frequently Asked Questions
Is 0.9% saline the same as 0.154 M?
Yes. 0.9% w/v means 9 g of NaCl per liter, and 9 ÷ 58.44 g/mol = 0.154 mol/L. The two labels describe the same concentration.
What is the molarity of half-normal saline?
Half-normal saline is 0.45% w/v, or 4.5 g/L, which is 4.5 ÷ 58.44 = 0.077 M — exactly half the molarity of normal saline.
Why does osmolarity matter for saline?
Cells swell or shrink when the surrounding fluid's particle concentration differs from blood. At about 308 mOsm/L, normal saline matches plasma closely enough to avoid damaging red blood cells.
How many mEq of sodium are in a liter of normal saline?
154 mEq. Since NaCl contributes one sodium ion per formula unit, the sodium concentration in mEq/L equals the molarity in mmol/L: 154.
